## Content

1.取100m新鮮植物或25mg乾燥植物 2.將葉片放入研缽中，加入液態氮磨成粉 3.加入1ml PR Buffer & 10ul -Mercaptoethanol 到研缽中，充分研磨。 4.將液體移到tube中，在75C 培養30mins (每10 mins混勻) 5.離心14000g ,4C ,10分鐘 ,將上清液移到新tube中。 6.加入1/2體積的isopropanol到樣本中，且混和均勻。 7.將 RZ column 放入 collection tube中 8.將sample mixture 移到RZ column(最多一次700,超過分兩次)，離心14000g ,4C ,30秒 9.去除下層液體 10.加入400ul W1 Buffer 11.離心14000g ,4C ,30秒 12.去除下層液體 13.加入600ul W2 Buffer 14.離心14000g ,4C ,30秒 15.去除下層液體 16.再次離心14000g , 4C ,2mins ,乾燥RE column 17.將RZ column放到新的tube中 18.加入50-90ul 的 Elution Buffer 到 column中心 19.室溫反應2 mins 20.離心14000g , 4C ,2mins 21.保存於-80C /測RNA濃度
