But the anomalies they found led the research team to focus on Lactobacillus, a probiotic that has been shown to be associated with AHR activity. It works mainly by metabolizing tryptophan in the diet, because microorganisms produce indole when metabolizing tryptophan, which happens to activate AHR. This is also the reason why AHR can still function when AHR activating gene is inactivated.

The researchers tried two ways to reproduce AHR inactivation in the tumor mouse model. One is to remove tryptophan from the daily diet. After losing tryptophan supply, the AhR activity in Tam decreased significantly, and more anti-tumor T cells infiltrated into the tumor tissue. This improvement disappeared when the researchers added indole to their diet.

Another way is to remove the intestinal bacteria in mice. After continuous administration of antibiotics against lactobacillus, the mice also had the changes when tryptophan was removed. These mice without Lactobacillus had significantly slower tumor growth and longer survival time than other control groups.

In addition, the study also tried to transplant lactobacillus with different indole production capacity into multiple groups of sterile mice. The results showed that the tumor growth rate of mice was significantly different about two weeks after transplantation, and the tumor burden of mice transplanted with indole producing strains was greater.

“Many studies have focused on the benefits of intestinal microorganisms for cancer, but we have found that probiotics may also have negative effects in some cases,” Dr. mcgaha said. He plans to collect stool samples from tumor patients before and after chemotherapy in the future to analyze the changes in the composition of intestinal flora, especially Lactobacillus, so as to confirm their correlation with treatment results.
